Phase 1/2 study of TP53 mutation-targeting therapy signals expansion of biotech-focused drug development strategy

LG Chem is accelerating its push into precision oncology with the launch of a U.S. clinical trial for a next-generation targeted cancer therapy aimed at patients with hard-to-treat genetic mutations for which no approved treatments currently exist.

The company announced on Monday that it has received clearance from the U.S. Food and Drug Administration (FDA) to initiate a Phase 1/2 clinical trial for its investigational cancer drug candidate LG00313112.

The compound was in-licensed in April through a global exclusive development and commercialization agreement with U.S.-based Frontier Medicines, marking an important addition to LG Chem’s oncology pipeline.

LG00313112 targets the TP53 Y220C mutation, a rare genetic alteration found in approximately 1–3% of all cancer patients. The mutation destabilizes the p53 tumor suppressor protein, weakening its ability to control abnormal cell growth.

LG Chem said the drug is designed to restore tumor-suppressing function by stabilizing the structurally altered p53 protein. It utilizes a covalent-binding mechanism that, according to the company, enhances both target affinity and durability of response, potentially improving therapeutic effectiveness.

Preclinical studies demonstrated antitumor activity at low doses as well as sustained efficacy over time. The candidate also showed activity in tumor models carrying concurrent KRAS mutations, suggesting broader potential applicability across complex cancer profiles.

Currently, there are no approved targeted therapies specifically addressing TP53-mutated cancers. Data from the U.S. National Cancer Institute (NCI) indicate that patients with TP53 mutations have a median survival of 29 months, significantly shorter than the 63 months observed in patients without the mutation.

To streamline development timelines, LG Chem has combined Phase 1 and Phase 2 into a single adaptive clinical protocol. Phase 1 will evaluate safety, tolerability, and recommended dosing in patients with advanced solid tumors—including ovarian, lung, and breast cancers—harboring the TP53 Y220C mutation. Phase 2 will then assess preliminary efficacy based on outcomes from the first stage.

Industry observers view the development as part of LG Chem’s broader strategy to strengthen its biotechnology and pharmaceutical portfolio beyond its traditional chemical business, with a focus on high-value precision medicine platforms.
