Dong-A ST unveiled encouraging preclinical findings for two next-generation Alzheimer's disease drug candidates at the Alzheimer's Association International Conference (AAIC) 2026, underscoring its strategy to develop disease-modifying therapies targeting multiple mechanisms of neurodegeneration.

The company presented poster sessions featuring DA-7505, a GPX4 activator, and DA-7503, a tau aggregation inhibitor, at the conference, one of the world's largest scientific gatherings dedicated to Alzheimer's disease research.

The presentations showcased the therapeutic potential of the two candidates in addressing distinct pathological pathways associated with Alzheimer's disease and other neurodegenerative disorders.

DA-7505 demonstrated significant neuroprotective effects by enhancing the activity of glutathione peroxidase 4 (GPX4), a key antioxidant enzyme that suppresses ferroptosis—an iron-dependent form of programmed cell death increasingly recognized as a major contributor to neurodegenerative diseases.

According to Dong-A ST, the compound effectively reduced lipid peroxidation and neuroinflammation while improving cognitive function in Alzheimer's disease animal models. The company also reported stronger anti-inflammatory activity than conventional reactive oxygen species (ROS)-targeting approaches.

The candidate is designed as a first-in-class small-molecule GPX4 positive allosteric modulator with high blood-brain barrier penetration, suggesting potential applications beyond Alzheimer's disease, including other neurodegenerative disorders characterized by ferroptosis.

The company also presented new findings on DA-7503, a small-molecule tau aggregation inhibitor currently undergoing Phase 1 clinical trials.

In preclinical studies, DA-7503 significantly improved cognitive and motor function in tauopathy animal models while reducing tau hyperphosphorylation, oligomer formation, and pathological tau accumulation throughout the brain.

The compound also showed enhanced therapeutic effects when administered in combination with lecanemab, an amyloid-beta-targeting antibody currently used for Alzheimer's disease treatment. The combination further reduced both amyloid-beta and tau pathology, supporting a dual-target therapeutic strategy that addresses two of the disease's primary pathological hallmarks.

Industry experts have increasingly emphasized the importance of multi-target approaches for Alzheimer's treatment as researchers seek therapies capable of slowing or modifying disease progression rather than simply alleviating symptoms.

The latest findings position Dong-A ST among companies exploring next-generation disease-modifying therapies that extend beyond conventional amyloid-focused treatment strategies.

"These results further validate the therapeutic potential of both DA-7503 and DA-7505 for neurodegenerative diseases," a Dong-A ST official said. "We will continue advancing innovative drug development targeting multiple disease mechanisms in Alzheimer's disease and tauopathies to address significant unmet medical needs."

The Alzheimer's Association International Conference (AAIC) is the world's largest annual scientific conference dedicated to Alzheimer's disease and dementia research, bringing together leading scientists, clinicians, and pharmaceutical companies to present the latest advances in the field.
